Description
This 18-plex assay contains 13 different arbovirus specific target proteins for detecting IgG antibodies to 10 different arboviruses, namely, Zika, Dengue (DENV1, DENV2, DENV3, DENV4), West Nile (WNV), Japanese encephalitis virus (JE), Yellow Fever (YFV), Tick Borne Encephalitis (TBEV), and Chikungunya (CHIKV). The test also includes five different internal controls to monitor each step of assay performance. This multiplex assay uses magnetic microspheres coupled with unique recombinant proteins specific for different viral targets to capture antibodies from human serum or plasma. The serum or plasma samples are mixed with the antigen-coated microspheres in wells of a 96-well plate and incubated for the antigen-antibody reaction to occur. The antigen-specific antibodies get immobilized on the microspheres, and unbound material is washed away after the incubation. The antigen captured human IgM antibodies are then detected using a Fluorescent Anti-human IgM-phycoerythrin (AHIgM-PE) reporter conjugate. After a final wash to remove the unbound conjugate, the microspheres are resuspended in the buffer and analyzed using Luminex® LX 200™ flow analyzer, or MAGPIX® or FLEXMAP 3D® instruments.
